Abstract: Akut miyokard infarktüs (AMI), kalbe giden kan dolaşımının aniden kesilmesi sonucunda, kalp kaslarının zarar görmesi ya da kalp kası hücrelerinin ölmesine sebep olabilmekte, bu da hastalarda sakatlık ya da ölüme yol açabilmektedir. Kalbin beslenememesi sonucu oluşan bu durumda, kalp kaslarının salgıladığı proteinler arasında Troponin-T de bulunmakta ve AMI başlangıcından itibaren ilk 1 saat içinde kandaki konsantrasyonu yaklaşık 2 katına çıkmaktadır. Bu çalışmada, plazma polimerizasyonu yöntemi kullanılarak kütle hassas kuvartz ayar çatalı (QTF) tabanlı bir Troponin-T sensörü geliştirilmesi amaçlanmıştır. Kuvartz ayar çatalı yüzeyi; n-heptilamin, pentilamin ve 1,2-diaminosiklohekzan ile farklı güç ve uygulama sürelerinde plazma polimerizasyonu yöntemi kullanılarak modifiye edilmiştir. Yüzey üzerinde elde edilen ince filmlerin temas açıları film kararlılığını belirlemek amacı ile 1 ay boyunca takip edilmiştir. Bunun yanı sıra elde edilen direnç ölçümlerinde direncin 0.01 M? seviyesinden 4.28 M? seviyesine kadar çıktığı gözlenmiştir.İnce film özellikleri, taramalı elektron mikroskobu (SEM), atomik kuvvet mikroskobu (AFM), X-ışını fotoelektron spektroskopisi (XPS) ve Fourier dönüşümlü kızılötesi spektroskopisi (FTIR) gibi görüntüleme ve spektroskopik yöntemler ile karakterize edilmiştir. QTF yüzeyine kaplanan filmler daha sonra farklı sürelerde gluteraldehit (GA) buharı ve çözeltisi ile etkileştirilerek kimyasal olarak aktive edilmiştir. Piezoelektrik özellik taşıyan QTF'lerin frekans değişimleri bu tüm değişim uygulanan süreçler öncesi ve sonrasında takip edilmiştir. Bu ölçümler kaplanan ince film hakkında bilgiler vermenin yanında, geliştirilmesi hedeflenen kütle hassas biyosensör için büyük önem taşımaktadır. Elde edilen sonuçlara göre, yüzey aktivasyonu yapılan QTF'lere farklı konsantrasyonlarda sığır serum albumin bağlanmış ve frekans değişimleri takip edilmiştir. Elde edilen verilerin, kütle hassas biyosensör platformlarının gelişimine öncülük etmesi beklenmektedir.
Acute myocardial infarction (AMI) may be the reason of irreversible myocardial cell damage or ischemic necrosis in case of a sudden loss in the blood supply to myocardium. This medical case can cause the death or disability of the patient. In case of ischemia, heart muscles release some proteins including Troponin-T. The concentration of this protein levels to twice of its initial concentration in blood within the first hour after heart attack. In this study, development of quartz tuning fork (QTF) based troponin-T biosensor by plasma polymerization method has been introduced. Surface modification of quartz tuning fork made by plasma polymerization method at diffrent discharge powers and exposure times with n-heptylamine, pentylamine and 1,2-diaminocyclohexane. Contact angle of the thin films deposited on the surface has been investigated for 30 days interval. In the resistivity measurements it has been seen that the initial resistivity has been increased from 0.01 M? to 4.28 M?. Thin film properties have been characterized by scanning electron microscopy (SEM), atomic force microscopy (AFM), X-Ray photoelectron spectroscopy (XPS) and fourier transform infrared spectroscopy (FTIR). These films were to exposed Glutheraldehyde (GA) in different mediums and exposure times in order to activate the surface. Frequency shifts of these QTFs were to be measured before and after glutheraldehyde activation. These measurements both gives important data on deposited thin fils and is very important for the development of the immunosensor. After the activation of the surfaces, QTFs ere to dipped into different concentrations of bovine serum albumin and the frequency shifts were investigated. These findings will be useful for the development of mass sensitive biosensor platforms.
