Abstract: Son dönemlerde doğadan esinlenerek yeni nesil biyouyumlu malzeme üretimi üzerine araştırmalar yapılmaktadır. Bu malzemelerin biyomedikal uygulamalarda kullanımı biyomateryaller, biyosensörler, fonksiyonel membranlar ve yapay doku iskeleleri gibi çeşitlilik göstermektedir. Bu çalışmada "doğal ve fonksiyonel" özellikleri olan biyokatalitik membran üretimi amaçlanmıştır. Bu hedef doğrultusunda, model altyapı proteini olarak seçilen sığır serum albümin ıslak kimya teknikleri ile amiloid forma dönüştürülmüştür. Glikoz oksidaz enzimi yapıdaki aktif biyolojik ajan olarak seçilmiştir. Biyokatalitik membran elektro-eğirme yöntemi ile "nano" ölçekte fiberlerin rastgele yığını şeklinde üretilmiştir. Tez kapsamında gerçekleştirilen çalışmalar, üretilen biyokatalitik membranın yapısındaki proteinin amiloid forma dönüştürülebildiğini, yapıdaki enzimin 0,7 µA akım şiddeti yarattığını ve 2547 U/m2 aktiviteye erişilebildiğini göstermiştir.
In the recent years, the researches on the production of new generation biocompatible materials are inspired by the nature. The use of these materials in biomedical applications vary as, biomaterials, biosensors, functional membranes and artificial tissue scaffolds. In this study, production of a biocatalytic membrane with "natural and functional" features, was intended. Towards this goal, bovine serum albumin was selected as the model infrastructure protein. Then, it had been transformed into amyloid form with the wet chemistry techniques. Glucose oxidase is selected as the active agent in the biological structure. Biocatalytic membrane was produced with electro-spinning method, in the form of random bundle of nanofibers. The results of the studies carried out within the scope of the thesis showed that, the protein used to produce biocatalytic membrane can be converted into amyloid form, the enzyme in the structure was created a current of 0.7 ?A and achived an activity of 2547 U/m2.
